The by far very best way is to study the in the fresh state. Everything else leads to minor results and includes artifacts.

I can only recomend you to read my paper from 1992 (Mycotaxon).
Preservation in Ascobolus is not different from any other ascomycete. Drying and placing in a tight plastic bag. Hoyer's liquid I remember a bit, it is killing the cells and perhaps also dissolves structures.
