Black disco on Larix decidua
Adam Polhorský, 31-05-2016 15:20
Hello everyone,

black apothecia(1,1mm) with remnants of a margin growing on a weathered dry stump of Larix decidua.

Asci: IKI-, 8-spored, Crozier+ (not sure)
Spores: 16-30 x 2,5-4um, with many small guttules, multicelled- up to 10.
Paraphyses: thread-like

In a field i thougt it would be something from Rhytismatales, but microscopy excluded my candidates.
Thanks for the advice

Adam

Re : Black disco on Larix decidua
My guess would have been Pragmpora amphibola, but that species has apos 0.3-0.8 mm, paraphyses with refractive intercellular substance, and perhaps shorter spores. Could you please correct your spore data, does it mean 16-30 or 30-36?

Re : Black disco on Larix decidua
U are right about the apo size, measured the largest one with the microscope - 1,1 mm.  but the spores are truly 16-30um (the free, released ones)
I looked at Pragmopora before but excluded it due to spore size/ septation, paraphyses are too thin 1,2um- maybe theres some content but i cannot see it w my setup, also the margin is different? atypical maybe... and substrate is different. Maybe within variation or closely related species?

Re : Black disco on Larix decidua
I think the oleaginous matter between the paraphyses (not within) is a valuable character, though not only in Pragmopora. I think you shoud see that if it is there. Best is to use water as medium.
I attach a drawing of mine and a photo by Luis Quijada.

Re : Black disco on Larix decidua
I dont see it Zotto, it looks easily observable but i see nothing, only asci and paraphyses nothing inbetween.
Maybe i can get it sequenced, but there´s no Pragmopora in genbank to compare with.

Re : Black disco on Larix decidua
It may be something else. Yes, it is not in GenBank, but I should get an inofficial sequence from our culture (ITS and perhaps LSU). So sequencing would be very important in this group.

i will let u know when i make progress with this collection.
